De novo Sequencing


MALDI MS/MS

Once a MALDI-MS spectra has been obtained an individual peptide ion can be selected for further fragmentation. Interpretation of the resultant product, or daughter ions, can be used to determine the peptide sequence and identify post-translational modifications.

The results from MS and MS/MS analysis can be combined and searched against databases of known proteins to identify those present in the sample. MS/MS data has the advantage over PMF in that it a higher level of confidence in the identity of the proteins can be attained and it is better able to identify multiple proteins within the same sample.

The sequence data alone can also be used to BLAST search sequence database to try and identify proteins which are not in the sequence database, such as those from species which have not had their genome sequenced.
